The easiest way to get glyphs in Diablo 4 is by completing Nightmare Dungeons. To unlock these, you will need some Nightmare Sigils. To get there, you will need to complete the Whispers the Dead quest where you're tasked with collecting ten Grim Favors. As this is a main story mission, you will eventually encounter it as you keep playing.
After completing the quest, you will unlock the Tree of Whispers as well as the Nightmare Sigils. Now, you can begin collecting the Sigils by participating in the Cathedral of Light Capstone Dungeon. Progress through the dungeons until you reach World Tier 3 Difficulty, and you will unlock Nightmare Dungeons. How to complete Nightmare Dungeons in Diablo 4
